Inner filter effect - a limiting factor in high-throughput analysis

The inner filter effect (IFE) is a common problem in all fluorescent measurements. It depends on the optical density of the sample and on the precise geometry of the sample illumination. There are two different effects present:

Primary IFE

In highly concentrated samples the excitation beam is absorbed only near the surface facing the ecvitation beam.

Secondary IFE

When excitation and emission spectra overlap, the light emitted can be reabsorbed by the rest of the samples.
