The inner filter effect (IFE) is a common problem in all fluorescent measurements. It depends on the optical density of the sample and on the precise geometry of the sample illumination. There are two different effects present:

Primary IFE

In highly concentrated samples the excitation beam is absorbed only near the surface facing the ecvitation beam.

Secondary IFE

When excitation and emission spectra overlap, the light emitted can be reabsorbed by the rest of the samples.

TINFE corrects the nonlinear response of emission spectra, including IFE, by taking at least two measurements at different settings, ensuring accurate and precise data with an extended range through advanced signal processing.

In our lab, determination of gold nanoparticles using fluorescent spectroscopy has been optimized by using TINFE method and software. We have seen a significant increase in productivity, by eliminating multiple dilution steps, thus making more analysis per day possible.
